Filler-binders for Direct Compression

A filler-binder provides more than bulk for a tablet. An effective filler-binder also possesses good flow and compactability, the two properties that make Direct Compression possible.

Directly compressible lactose

SuperTab® spray dried lactose

Spray dried lactose is specifically designed for Direct Compression applications, and its spherical structure and high flow properties, together with its low sensitivity to compaction speed, make it the ideal filler-binder for high speed tableting applications.

SuperTab® 11SD is the class leading grade of spray dried lactose used by the pharmaceutical industry. SuperTab® 14SD is spray dried lactose with high compaction properties conferred by optimisation of the amorphous content and the size of the primary lactose particles.

SuperTab® anhydrous lactose

Anhydrous lactose is the form of lactose with the lowest water content, and yet it exhibits very low hygroscopicity, making it the material of choice when very low total water content is required. Its brittle nature confers low strain rate sensitivity and low lubricant sensitivity thereby minimising the risks of product failure on scale-up from laboratory to factory. Anhydrous lactose is also the lactose of choice for Dry Granulation applications because of its ability to recompress well after the initial densification process.

SuperTab® AN grades are anhydrous lactose. SuperTab® 21AN forms good plugs at low forces making it a good choice for hard gelatin capsule formulations. SuperTab® 22AN is processed with a particle size distribution optimised for good flow.

SuperTab® granulated lactose

SuperTab® 30GR is lactose monohydrate processed by fluid bed agglomeration into free flowing compressible granules. It is the perfect all round form of Direct Compression lactose exhibiting good flow and compaction, very low hygroscopicity and excellent mixing potential. Tablets made form SuperTab® 30GR exhibit excellent storage stability.

Microcrystalline cellulose

Additionally we offer Pharmacel 102 microcrystalline cellulose for DC applications. Pharmacel is microcrystalline cellulose. It confers exceptional compactability on Direct Compression tablet formulations and the resulting tablets are generally readily disintegrated. For Direct Compression the coarser Pharmacel 102 grade may be preferred.

Microcrystalline cellulose is also extensively used in Dry Granulation because of its high compactability. In this application the finer Pharmacel 101 grade may be preferred.
